    Independence Day for the “Old Lady? A Natural Experiment on the Implications of Central Bank Independence

    Central bank independence is widely thought be a sine qua non of a credible commitment to price stability. The surprise decision by the UK government to grant operational independence to the Bank of England in 1997 affords us a natural experiment with which to gauge the impact on the yield curve from the adoption of central bank independence. We document the extent to which the decision to grant independence was ‘news?and illustrate that the reduction in medium and long term nominal interest rates was some 50 basis points, which we show to be consistent with a sharp increase in policymaker’s aversion to inflation deviations from target. We suggest therefore central bank independence represents one of the clearest signals available to elected politicians about their preferences on the control of inflation.Central bank independence; preferences; yield curve.

    Content and Concept: An Examination of Transcendental Empiricism

    In this dissertation, I critically examine the philosophy of transcendental empiricism. Transcendental empiricism is, among other things, a philosophy of mental content. It attempts to dissolve an epistemological dilemma of mental content by splitting the difference between two diametrically opposed accounts of content. John McDowell\u27s minimal empiricism and Richard Gaskin\u27s minimalist empiricism are two versions of transcendental empiricism. Transcendental empiricism itself originates with McDowell\u27s work. This dissertation is divided into five parts. First, in the Introduction, I state the Wittgensteinian metaphilosophical orientation of transcendental empiricism. This metaphilosophical approach provides a plateau upon which much of the rest of this work may be examined. Second, I offer a detailed description of McDowell\u27s minimal empiricism. Third, I critique Gaskin\u27s critique and modification of McDowell\u27s minimal empiricism. I argue that (1) Gaskin\u27s critiques are faulty and that (2) Gaskin\u27s minimalist empiricism is very dubious. Fourth, I scrutinize the alleged credentials of McDowell\u27s minimal empiricism. I argue that McDowell\u27s version of linguistic idealism is problematic. I then comment on a recent dialogue between transcendental empiricism and Hubert Dreyfus\u27s phenomenology. The dialogue culminates with Dreyfus\u27s accusation of the Myth of the Mental. I argue that this accusation is correct in which case McDowell\u27s direct realism is problematic. I conclude that minimal empiricism does not dissolve the dilemma of mental content. Finally, I argue that Tyler Burge successfully undermines the doctrine of disjunctivism, but disjunctivism is crucial for transcendental empiricism. Ultimately, however, I aim to show that transcendental empiricism is an attractive alternative to philosophies of mental content

NCL's reliance on the redox active amino acid cysteine and β-mercaptamine derivatives suggests a potential role for electrochemistry.     Investigation of acyl transfer auxiliary-assisted glycoconjugation for glycoprotein semi-synthesis

    Homogeneous glycoprotein syntheses have become possible in the last decade due to advances in chemical ligation strategies, particularly Native Chemical Ligation (NCL). For native glycoproteins this still requires laborious and technically challenging syntheses of glycopeptide components, combined with multi-segment ligation reactions. Here we explore new reactions between sugar-linked acyl transfer auxiliaries and peptide thioesters. We show that native glycoproteins are difficult to produce using this approach but various related analogues are accessible. The results show that site-specific neoglycoconjugation is a viable route to simply glycosylated proteins, which may be extended using well-documented enzymatic processes

    Effects of Corn Oil and Benzyl Acetate on Number and Size of Azaserine-Induced Foci in the Pancreas of LEW and F344 Rats

    The response of LEW and F344 strain rats to the pancreatic carcinogen azaserine was compared using the size and number of azaserine-induced acidophilic acinar cell foci and nodules as parameters in a 4-month experiment. A second experiment compared the effect of corn oil intake by gavage and dietary routes on the growth of azaserine-induced pancreatic lesions in LEW rats. A third experiment tested the activity of benzyl acetate in regard to its ability to induce acinar cell foci or to promote the growth of such foci in azaserine-treated rats. The results showed that equivalent doses of azaserine induce two to seven times more foci in LEW than in F344 rats, and that LEW rats have a higher incidence of spontaneous foci than F344 rats. Azaserine-treated LEW rats that were given 5 mL corn oil/kg body weight 5 days per week by gavage developed more acinar cell foci than rats fed a basal diet (chow). Addition of an equivalent amount of corn oil to chow had a similar effect of enhancing the development of foci. Rats of neither strain developed acinar cell foci when benzyl acetate was given by gavage or in the diet nor was there evidence that benzyl acetate has a significant effect on the development of foci in azaserine-treated rats. These studies also demonstrate that the azaserine/rat model of pancreatic carcinogenesis which was developed in LEW rats can be adapted for use with F344 rats

    Disruption of membrane cholesterol organization impairs the activity of PIEZO1 channel clusters

    The human mechanosensitive ion channel PIEZO1 is gated by membrane tension and regulates essential biological processes such as vascular development and erythrocyte volume homeostasis. Currently, little is known about PIEZO1 plasma membrane localization and organization. Using a PIEZO1-GFP fusion protein, we investigated whether cholesterol enrichment or depletion by methyl-β-cyclodextrin (MBCD) and disruption of membrane cholesterol organization by dynasore affects PIEZO1-GFP’s response to mechanical force. Electrophysiological recordings in the cell-attached configuration revealed that MBCD caused a rightward shift in the PIEZO1-GFP pressure–response curve, increased channel latency in response to mechanical stimuli, and markedly slowed channel inactivation. The same effects were seen in native PIEZO1 in N2A cells. STORM superresolution imaging revealed that, at the nanoscale, PIEZO1-GFP channels in the membrane associate as clusters sensitive to membrane manipulation. Both cluster distribution and diffusion rates were affected by treatment with MBCD (5 mM). Supplementation of polyunsaturated fatty acids appeared to sensitize the PIEZO1-GFP response to applied pressure. Together, our results indicate that PIEZO1 function is directly dependent on the membrane composition and lateral organization of membrane cholesterol domains, which coordinate the activity of clustered PIEZO1 channels

    An ever-growing number and length of documents, number and depth of topics covered by legislation, and ever new phrases and their slowly changing meaning, these are all contributing factors that make policy analysis more and more complex. As implication, human policy analysts and policy developers face increasing entanglement of both content and semantical levels. To overcome several of these issues, JRC has developed a central pilot tool called AI-KEAPA to support policy analysis and development in any domain. Recent developments in big data, machine learning and especially in natural language processing allow converting unfathomable complexity of many hundreds of thousands of documents into a normalised high-dimensional vector space preserving the knowledge. Unstructured text in document corpora and big data sources, until recently considered just an archive, is quickly becoming core source of analytical information using text mining methods to extract qualitative and quantitative data. Semantic analysis allows us to extract better information for policy analysis from metadata titles and abstracts than from the structured human-entered descriptions. This digital assistant allows document search and extraction over many different sources, discovery of phrase meaning, context and temporal development. It can recommend most relevant documents including their semantic and temporal interdependencies. But most importantly, it helps bursting knowledge bubbles and fast-learning new domains. This way we hope to mainstream artificial intelligence into policy support. The tool is now fit for purpose. It was thoroughly tested in real-life conditions for about two years mainly in the area of legislative impact assessments for policy formulation, and other domains such as large data infrastructure analysis, agri-environmental measures or natural disasters, some of which are detailed in this document. This approach boosts the strategic JRC focus on application of scientific analysis and development.     Evidence for language transfer leading to a perceptual advantage for non-native listeners

    Phonological transfer from the native language is a common problem for non-native speakers that has repeatedly been shown to result in perceptual deficits vis-a-vis native speakers. It was hypothesized, however, that transfer could help, rather than hurt, if it resulted in a beneficial bias. Due to differences in pronunciation norms between Korean and English, Koreans in the U.S. were predicted to be better than Americans at perceiving unreleased stops--not only in their native language (Korean) but also in their non-native language (English). In three experiments, Koreans were found to be significantly more accurate than Americans at identifying unreleased stops in Korean, at identifying unreleased stops in English, and at discriminating between the presence and absence of an unreleased stop in English. Taken together, these results suggest that cross-linguistic transfer is capable of boosting speech perception by non-natives beyond native levels
